چکیده

Background: Age is the most important risk factor in degenerative osteoarthritis (OA) and associated with accumulation of senescent cells that contribute to functional decline joint. We previously demonstrated extracellular vesicles (EVs) from mesenchymal stromal (MSCs) largely mediate therapeutic effect parental OA. Objectives: Here, we assessed impact senescence on characteristics EVs adipose tissue-derived MSCs (ASC-EVs) their properties an vitro model OA Methods: ASCs were induced using 25µM etoposide for 24 hours. Senescence was by quantifying proliferation rate, SA-βGal activity, nuclear γH2AX foci number, phalloidin staining expression cyclin dependent kinase inhibitors (CDKI) (RT-qPCR). ASC-EVs isolated differential ultracentrifugation characterized size, concentration, total protein content, structure (cryo-TEM) immunophenotype. In used chondrocytes patients, which stimulated IL1β 48h before culture or 7 days. Expression chondrocytic inflammatory markers quantified RT-qPCR SASP factors ELISA supernatants. Results: Senescence-induced experienced growth arrest increase staining, p21 CDKI expression, foci, stress fibers several (IL6, IL8, MMP3) confirming main features. Senescent produced 4-fold more than healthy larger. vitro, both decreased fibrotic ( type III COLLAGEN ), catabolic hypertrophic MMP3, MMP13, AP ) increased COX2 chondrocytes. By contrast, IL6 while highly . Looking at role chondrocytes, found out able AGG II they AP, X COLLAGEN, HMOX1 Finally, number positive but did not IL1β-induced Conclusion: Our results indicated a chondroprotective ASC-EVs, independently state suggested might act through different mechanisms ASCs, warrants further investigation Disclosure Interests: Jérémy Boulestreau: None declared, Marie Maumus Employee of: Bauerfeind France, Pauline Rozier: Christian Jorgensen Shareholder Medxcell sciences, Consultant Daniele Noel sciences

Extracellular Vesicles from Adipose-Derived Mesenchymal Stem Cells Downregulate Senescence Features in Osteoarthritic Osteoblasts

Osteoarthritis (OA) affects all articular tissues leading to pain and disability. The dysregulation of bone metabolism may contribute to the progression of this condition. Adipose-derived mesenchymal stem cells (ASC) are attractive candidates in the search of novel strategies for OA treatment and exert anti-inflammatory and cytoprotective effects on cartilage. Chronic inflammation in OA is a re...

Comparative proteomic analysis of extracellular vesicles isolated from porcine adipose tissue-derived mesenchymal stem/stromal cells

Extracellular vesicles (EVs) isolated from mesenchymal stem/stromal cells (MSCs) contribute to recovery of damaged tissue. We have previously shown that porcine MSC-derived EVs transport mRNA and miRNA capable of modulating cellular pathways in recipient cells. To identify candidate factors that contribute to the therapeutic effects of porcine MSC-derived EVs, we characterized their protein car...

Chromatographically isolated CD63+CD81+ extracellular vesicles from mesenchymal stromal cells rescue cognitive impairments after TBI.

Extracellular vesicles (EVs) secreted by cells present an attractive strategy for developing new therapies, but progress in the field is limited by several issues: The quality of the EVs varies with the type and physiological status of the producer cells; protocols used to isolate the EVs are difficult to scale up; and assays for efficacy are difficult to develop. In the present report, we have...

Extracellular vesicles from bone marrow mesenchymal stem/stromal cells transport tumor regulatory microRNA, proteins, and metabolites

Human mesenchymal stem/stromal cells (hMSCs) have been shown to support breast cancer cell proliferation and metastasis, partly through their secretome. hMSCs have a remarkable ability to survive for long periods under stress, and their secretome is tumor supportive. In this study, we have characterized the cargo of extracellular vesicular (EV) fraction (that is in the size range of 40-150nm) o...
